## How the Test Works

The Reaction Mechanism

The test utilizes a synthetic chromogenic substrate that is cleaved by an enzyme activated during the LAL cascade. When endotoxin is present, it triggers a series of enzymatic reactions that ultimately lead to the release of p-nitroaniline (pNA) from the chromogenic substrate. The released pNA produces a yellow color that can be measured spectrophotometrically at 405 nm.

The key advantage of this method lies in its kinetic nature. Instead of measuring the endpoint color development, the test monitors the rate of color formation over time. The time required to reach a predetermined absorbance threshold (onset time) is inversely proportional to the endotoxin concentration in the sample.
